Moderna has delivered one of the most important results in its history since the development of its Covid-19 vaccine, reporting that its personalized mRNA cancer vaccine significantly reduced the risk of melanoma returning or spreading when used together with Merck’s blockbuster immunotherapy drug Keytruda. The breakthrough has revived hopes that mRNA technology could become a major new weapon against cancer—and has dramatically changed investors’ outlook on Moderna.
The experimental treatment, known as Intismeran, is designed individually for each patient. Doctors analyze genetic mutations from a patient’s tumor and use that information to create a customized mRNA vaccine intended to train the immune system to recognize and attack cancer cells. Unlike conventional vaccines, which generally prevent infectious diseases, this treatment is designed to help the body fight cancer after it has already developed.
In a late-stage clinical trial involving 1,137 patients with high-risk melanoma that had been surgically removed, the combination of Intismeran and Keytruda produced a statistically and clinically significant improvement compared with Keytruda alone. Earlier studies had shown a roughly 44% reduction in the risk of recurrence or death, helping establish the rationale for the larger trial. No major new safety concerns emerged from the latest study.
The result is particularly significant because it marks the first time a personalized mRNA cancer vaccine has demonstrated such a strong benefit in a large late-stage trial when added to an established checkpoint-inhibitor therapy. Merck and Moderna are now discussing the findings with regulators, and analysts believe the therapy could potentially receive approval as early as 2027 if the remaining data and regulatory review are favorable.
For Moderna, the timing could hardly be more important.
The company became one of the world’s most prominent biotechnology businesses during the pandemic, but Covid vaccine sales later collapsed as global demand declined. Moderna subsequently suffered heavy financial losses, falling investor confidence and questions about whether its enormous mRNA research pipeline could produce another commercially important product.
The melanoma results immediately changed that narrative. Moderna shares surged dramatically after the announcement, at one point rising more than 100% and adding tens of billions of dollars to the company’s market value. Investors are now betting that the vaccine could become not merely a melanoma treatment but evidence that Moderna’s underlying technology can work across multiple cancers.
That broader potential is crucial. Moderna and Merck are already testing personalized cancer vaccines in non-small-cell lung cancer, bladder cancer and kidney cancer, among other tumors. If the approach proves effective across several solid cancers, analysts believe it could generate billions of dollars in annual revenue. Some estimates suggest melanoma alone could eventually produce approximately $3 billion in annual sales by 2035.
Still, major uncertainties remain.
Detailed results from the late-stage trial have not yet been fully presented, and researchers will want to examine overall survival, long-term safety and whether the treatment’s benefits persist over time. Manufacturing is another challenge because each vaccine must be individually designed and produced for a specific patient, making the process more complex than manufacturing traditional medicines at large scale.
Cost will also matter. Moderna has not announced a price, although the company expects the treatment to cost less than highly customized therapies such as CAR-T cell treatments.
For cancer research, however, the importance extends beyond Moderna’s financial recovery. The results provide some of the strongest evidence yet that mRNA technology can be used not only to prevent infectious diseases, but also to direct the immune system against cancer.
That possibility could represent a major shift in oncology. Instead of treating every patient with exactly the same therapy, future cancer treatment may increasingly combine powerful immunotherapy drugs with vaccines built specifically around the genetic characteristics of an individual person’s tumor.
For Moderna, a company whose identity became inseparable from Covid, the melanoma breakthrough may finally demonstrate what it has argued for more than a decade: that mRNA is not simply a pandemic technology, but a platform capable of transforming medicine far beyond infectious disease.
